    Single-cell RNA sequencing identifies distinct mouse medial ganglionic eminence cell types.

    Many subtypes of cortical interneurons (CINs) are found in adult mouse cortices, but the mechanism generating their diversity remains elusive. We performed single-cell RNA sequencing on the mouse embryonic medial ganglionic eminence (MGE), the major birthplace for CINs, and on MGE-like cells differentiated from embryonic stem cells. Two distinct cell types were identified as proliferating neural progenitors and immature neurons, both of which comprised sub-populations. Although lineage development of MGE progenitors was reconstructed and immature neurons were characterized as GABAergic, cells that might correspond to precursors of different CINs were not identified. A few non-neuronal cell types were detected, including microglia. In vitro MGE-like cells resembled bona fide MGE cells but expressed lower levels of Foxg1 and Epha4. Together, our data provide detailed understanding of the embryonic MGE developmental program and suggest how CINs are specified

So, association rule hiding methods are applied to evade the hazard of delicate information misuse. Various kinds of investigation already prepared on association rule protecting. However, maximum of them concentrate on introducing methods with a limited view outcome for inactive databases (with only existing information), while presently the researchers facing the problem with continuous information. Moreover, in the era of big data, this is essential to optimize current systems to be suited concerning the big data. This paper proposes the framework is achieving the data anonymization by using fuzzy logic by supporting big data mining. The fuzzy logic grouping the sensitivity of the association rules with a suitable association level. Moreover, parallelization methods which are inserted in the present framework will support fast data mining process

    Flotation Studies on Low Grade Magnesite Deposits from Sujikonda near Daroji Bellary district, Karnataka State

    Magnesite, an important basic refractory raw material, has been in short supply in recent years. To find an alte-rnative to high costlow boron containing sea water magne-site, users have been looking more towards sources of a natural magnesite. Magnesite ( MgCO3 ) is commonly found in granular, compact earthy masses occurring as amorphous or crystalline deposits. The amorphous type of deposits are the most common variety found as veins in serpentine rocks. The low grade ores produced do not meet the chemical purity required by the consuming industries (Table-1). The low grade ores have to be upgraded by physical benefi-ciation methods to render them usable, besides, conserving high grade reserves of magnesite. in view of the proposed Vijayanagara steel plant in Toranagallu, the occurrence of Sujikonda magnesite deposit close to the steel plant attains greater significance. The magnesite deposit of Sujikonda area is of low grade with MgO = 29:60 wt.% ; Ca0 = 5.61 wt. %; Si02 = 3.84 wt. %; and R2 03 = 31.04 wt. %.     Beneficial Image Preprocessing by Contrast Enhancement Techniquefor SEM Images

    In this paper a morphological filtering algorithm using an exposure thresholding and measures of central tendency hasbeen proposed for solving the low contrast of Scanning Electron Microscopic (SEM) images of composite materials foraccurate Filler Content Estimation. SEM image of a composite material comprises visible morphological structures likefillers such as silica nanoparticles. The SEM image analysis via segmentation will assist in the study of distribution of thesestructures. The estimation of the filler content is more accurate only when the SEM images have proper contrast for analysisif not the results lead to less accuracy. To overcome this drawback, we have proposed a preprocessing technique to increasethe contrast of SEM images. So that the preprocessed image can be used for post processing namely segmentation and hencethe error is less for filler content estimation. We introduced the transformations using morphological processing to extractthe bright and darker features of the images. The optimum threshold value is determined by the image exposure. A detailedcomparative analysis with other existing techniques has been performed to prove the superior performance of the proposedmethod

    The timing and appropriateness of surgical treatment of sigmoid diverticular disease remain a topic of controversy. We have reviewed the current literature on this topic, focusing on issues related to the indications and types of surgery. Current evidence would suggest that elective surgery for diverticulitis can be avoided in patients with uncomplicated disease, regardless of the number of recurrent episodes. Furthermore, the need for elective surgey should not be influenced by the age of the patient. Operation should be undertaken in patients with severe attacks, as determined by their clinical and radiological evaluation

    Oncogenic ERBB3 Mutations in Human Cancers

    SummaryThe human epidermal growth factor receptor (HER) family of tyrosine kinases is deregulated in multiple cancers either through amplification, overexpression, or mutation. ERBB3/HER3, the only member with an impaired kinase domain, although amplified or overexpressed in some cancers, has not been reported to carry oncogenic mutations. Here, we report the identification of ERBB3 somatic mutations in ∼11% of colon and gastric cancers. We found that the ERBB3 mutants transformed colonic and breast epithelial cells in a ligand-independent manner. However, the mutant ERBB3 oncogenic activity was dependent on kinase-active ERBB2. Furthermore, we found that anti-ERBB antibodies and small molecule inhibitors effectively blocked mutant ERBB3-mediated oncogenic signaling and disease progression in vivo

    Deep RNA sequencing analysis of readthrough gene fusions in human prostate adenocarcinoma and reference samples

    <p>Abstract</p> <p>Background</p> <p>Readthrough fusions across adjacent genes in the genome, or transcription-induced chimeras (TICs), have been estimated using expressed sequence tag (EST) libraries to involve 4-6% of all genes. Deep transcriptional sequencing (RNA-Seq) now makes it possible to study the occurrence and expression levels of TICs in individual samples across the genome.</p> <p>Methods</p> <p>We performed single-end RNA-Seq on three human prostate adenocarcinoma samples and their corresponding normal tissues, as well as brain and universal reference samples. We developed two bioinformatics methods to specifically identify TIC events: a targeted alignment method using artificial exon-exon junctions within 200,000 bp from adjacent genes, and genomic alignment allowing splicing within individual reads. We performed further experimental verification and characterization of selected TIC and fusion events using quantitative RT-PCR and comparative genomic hybridization microarrays.</p> <p>Results</p> <p>Targeted alignment against artificial exon-exon junctions yielded 339 distinct TIC events, including 32 gene pairs with multiple isoforms. The false discovery rate was estimated to be 1.5%. Spliced alignment to the genome was less sensitive, finding only 18% of those found by targeted alignment in 33-nt reads and 59% of those in 50-nt reads. However, spliced alignment revealed 30 cases of TICs with intervening exons, in addition to distant inversions, scrambled genes, and translocations. Our findings increase the catalog of observed TIC gene pairs by 66%.</p> <p>We verified 6 of 6 predicted TICs in all prostate samples, and 2 of 5 predicted novel distant gene fusions, both private events among 54 prostate tumor samples tested. Expression of TICs correlates with that of the upstream gene, which can explain the prostate-specific pattern of some TIC events and the restriction of the <it>SLC45A3-ELK4 </it>e4-e2 TIC to <it>ERG</it>-negative prostate samples, as confirmed in 20 matched prostate tumor and normal samples and 9 lung cancer cell lines.</p> <p>Conclusions</p> <p>Deep transcriptional sequencing and analysis with targeted and spliced alignment methods can effectively identify TIC events across the genome in individual tissues. Prostate and reference samples exhibit a wide range of TIC events, involving more genes than estimated previously using ESTs. Tissue specificity of TIC events is correlated with expression patterns of the upstream gene. Some TIC events, such as <it>MSMB-NCOA4</it>, may play functional roles in cancer.</p

    Establishment and cryptic transmission of Zika virus in Brazil and the Americas

    Transmission of Zika virus (ZIKV) in the Americas was first confirmed in May 2015 in northeast Brazil1. Brazil has had the highest number of reported ZIKV cases worldwide (more than 200,000 by 24 December 20162) and the most cases associated with microcephaly and other birth defects (2,366 confirmed by 31 December 20162). Since the initial detection of ZIKV in Brazil, more than 45 countries in the Americas have reported local ZIKV transmission, with 24 of these reporting severe ZIKV-associated disease3. However, the origin and epidemic history of ZIKV in Brazil and the Americas remain poorly understood, despite the value of this information for interpreting observed trends in reported microcephaly. Here we address this issue by generating 54 complete or partial ZIKV genomes, mostly from Brazil, and reporting data generated by a mobile genomics laboratory that travelled across northeast Brazil in 2016. One sequence represents the earliest confirmed ZIKV infection in Brazil. Analyses of viral genomes with ecological and epidemiological data yield an estimate that ZIKV was present in northeast Brazil by February 2014 and is likely to have disseminated from there, nationally and internationally, before the first detection of ZIKV in the Americas. Estimated dates for the international spread of ZIKV from Brazil indicate the duration of pre-detection cryptic transmission in recipient regions. The role of northeast Brazil in the establishment of ZIKV in the Americas is further supported by geographic analysis of ZIKV transmission potential and by estimates of the basic reproduction number of the virus
